Sorry to hear that you're unable to interact with the Uffentune Mastery Challenge shrine.

I've contacted our specialists about this, and we noticed that before you're able to activate the new set of Mastery Challenges, you need to have completed the narrative quests from part 1 and have found the Eye of Odin in Freya's vault.
