I have a Tuwai radar at 300 encounters. I have my character parked at a shrine and currently all the overworld tuwai are normal. Can I just despawn them (either by leaving, closing the game or waiting) to get fresh chances at the luma or do I have to encounter them to get "new" spawns?

It depends. Single encounter that are lumas appear as luma right away. But sometimes, you see a normal tem, and when you encounter it it's a double encounter, with the other tem being luma. Meaning there can still be a luma even if it doesn't look like one.
