If you log out of the account and log back in, does that allow you to spend the orbs?

8-card boosters from the store are not guaranteed to have a planeswalker - they're just distributed by rarity in those. The boosters used in draft and sealed each have a planeswalker, similar to printed boosters.

Does this happen immediately when trying to find a match or only after you wait in the queue for some time?

it looks like if you get all 25 orbs before you play with a different colour deck, you can no longer unlock the coloured orbs, even if you play with a different coloured deck.

Yeah, we have this in our backlog to fix. There's a workaround - if you log out and back in from the options menu, it will let you spend the orbs once you've played with the relevant deck.
