You can be on the character selection screen, playing an alt, or playing your characters with issues.

Now that we have launched, are you experiencing the same issue or did it resolve when the servers were stood up and stabilized?
You definitely have to be online, in-game. I am waiting to hear if you have to be on that specific character.
They will be able to resolve your lost quests from the backend.
